K-HELP MAGE-A4 (also referred to as H/K-HELP-MAGE-A4 or MAGE-A4-H/K-HELP) is an artificially synthesized helper/killer-hybrid epitope long peptide vaccine derived from the melanoma-associated antigen A4 (MAGE-A4), a cancer-testis antigen overexpressed in various cancers. This peptide vaccine is designed to induce both cellular and humoral immune responses against tumors expressing the MAGE-A4 antigen. Mechanistically, vaccination with K-HELP MAGE-A4 stimulates Th1 and Tc1 immune responses and promotes the production of complement-fixing IgG antibodies specific for the MAGE-A4 antigen. Clinical reports indicate that this approach can lead to tumor regression and decreased tumor markers in patients with metastatic cancer[5]. The therapy has been studied in combination with adjuvants such as OK432 and Montanide ISA-51.
