K01-162 (also referred to as K162) is a small molecule research compound designed to target amyloid-beta (Aβ) peptides, particularly the Aβ42 isoform, for the potential treatment of Alzheimer's disease. It acts by binding to and destabilizing amyloid-beta oligomers (AβO) with an EC50 of 80 nM. Mechanistically, it stabilizes the hydrophobic core I of the Aβ42 peptide (residues 17–21) into an α-helical conformation, which inhibits the formation of toxic fibrils and neutralizes their neurotoxicity. K01-162 is brain-penetrant and has demonstrated the ability to reduce brain amyloid burden in vivo in both fibrillar and non-fibrillar forms. Currently, it is primarily used as a research reagent and is sold by various chemical suppliers for laboratory use.
