K1-70 is a human monoclonal autoantibody that specifically targets and binds with high affinity to the thyroid stimulating hormone receptor (TSHR). By blocking ligand binding, it prevents stimulation of the TSHR by both endogenous thyroid stimulating hormone (TSH) and pathogenic autoantibodies found in Graves' disease. This antagonism leads to reduced thyroid hormone production, resulting in hypothyroid effects. K1-70 has been developed primarily for the treatment of Graves' disease and associated Graves' orbitopathy (GO), conditions characterized by hyperthyroidism and eye symptoms due to autoimmune activation of TSHR. In phase I clinical trials, K1-70 was well tolerated with no significant immunogenicity or serious adverse events observed. The drug demonstrated expected pharmacodynamic effects—lowering fT3 and fT4 levels while increasing TSH—and showed clinically meaningful improvements in both systemic symptoms of Graves’ disease and ocular manifestations of GO[1][2][4][5].
