K1.1d-mRNA-LNP is a preclinical mRNA-based therapeutic candidate developed for the treatment of human hepatocellular carcinoma (HCC), the most common type of primary liver cancer in adults and a leading cause of cancer-related deaths worldwide[1][2][6]. The drug consists of a truncated version (K1.1d) of an mRNA sequence encoding a protein or peptide with anti-tumor activity, encapsulated within specially designed lipid nanoparticles (LNPs) to facilitate systemic delivery and tumor targeting[2][6]. In mouse models bearing orthotopic human HCC tumors, systemic administration of K1.1d-mRNA-LNP resulted in efficient delivery to tumor sites and demonstrated significant dose-dependent anti-tumor activity with good tolerability[2][6]. The mechanism is based on mRNA-induced expression within tumor cells, potentially triggering direct cytotoxic effects or enhancing immune recognition against the cancer cells; however, the precise molecular target encoded by K1.1d has not been publicly disclosed[6]. Sunshine Biopharma is actively optimizing dosing regimens for this truncated version due to its promising efficacy at lower doses compared to full-length constructs[2][6].
