Drug intelligence / Profile preview
K101.1 (GRP94 antibody) is an investigational fully human monoclonal antibody developed by Hauul Bio and Kookmin University that specifically targets cell surface glucose-regulated protein 94 (GRP94, also known as endoplasmin or HSP90B1). GRP94 is a molecular chaperone belonging to the heat shock protein 90 (HSP90) family that is overexpressed on the surface of various cancer cells, including colorectal cancer (CRC) and gastric cancer (GC), where it plays a key role in tumor cell survival, growth, angiogenesis, and metastasis. Isolated using phage display technology, K101.1 binds to a specific linear epitope (Glu26-Ala34) on cell surface GRP94, inducing rapid internalization and downregulation of GRP94 expression. Preclinical studies have demonstrated that K101.1 effectively inhibits tumor growth and angiogenesis in vivo in both colorectal and gastric cancer xenograft models without causing severe systemic toxicity, highlighting its potential as a targeted therapeutic agent.
