K101CAR T cells are a form of **chimeric antigen receptor (CAR) T cell therapy** specifically engineered to target the CA125 extracellular repeat domain of the MUC16 glycoprotein, which is commonly expressed on ovarian cancer cells. The construct was developed by identifying and adapting high-affinity antibodies (notably clone K101) into a CAR format, resulting in T cells that show high potency and specificity for the CA125 domain. Preclinical studies demonstrated **significant efficacy of K101CAR T cells against cell lines and patient-derived tumor models of ovarian cancer, both in vitro and in vivo**. The function of these CAR T cells is not impaired by the presence of soluble CA125, which addresses a potential mechanism of immune evasion. The drug is being explored primarily for the treatment of MUC16-positive malignancies, with a focus on ovarian cancer[1].
