Overview
K3 CpG ODN is a synthetic class B cytosine-phosphate-guanine oligodeoxynucleotide (CpG ODN) designed as an immunotherapy adjuvant. It functions as a direct agonist of Toll-like receptor 9 (TLR9), resulting in strong activation of innate immunity, induction of a Th1-type adaptive immune response, enhanced cytotoxic T lymphocyte activity, and increased antibody production. K3 has been investigated primarily as an immune adjuvant for cancer immunotherapy (especially lung cancer) and as a vaccine adjuvant. K3 CpG ODN triggers rapid production of type I and II interferons and various cytokines, and promotes activation and maturation of dendritic cells and B cells. It has demonstrated safety and tolerability in phase I human clinical trials via subcutaneous, intravenous, and intraperitoneal routes[1][2][3][4][7][5]. Modified nanoparticulate forms (K3-SPG: K3 CpG ODN wrapped by schizophyllan) enhance delivery and immune induction[5][1].
