K658 is an adeno-associated virus (AAV) vector-based gene therapy developed by Chengdu Origen Biotechnology for the treatment of wet age-related macular degeneration (wet AMD). The therapy utilizes an AAV vector to deliver a gene encoding a vascular endothelial growth factor (VEGF) receptor fusion protein directly to retinal cells. Upon expression, this fusion protein acts as a decoy receptor that binds to and neutralizes VEGF, thereby inhibiting the pathological angiogenesis and vascular leakage that drive the progression of wet AMD. By enabling the continuous intraocular production of the therapeutic protein, K658 aims to provide a long-lasting treatment effect from a single administration, potentially overcoming the limitations of current anti-VEGF therapies that require frequent intravitreal injections.
