Overview

K757 is an investigational oral small-molecule GPR40 agonist developed by Kallyope, Inc. It belongs to a class of nutrient receptor agonists designed to target the gut-brain axis. By stimulating gut enteroendocrine cells, K757 triggers the secretion of satiety hormones such as glucagon-like peptide-1 (GLP-1), peptide YY (PYY), and cholecystokinin (CCK), mimicking the hormonal effects of bariatric surgery to suppress appetite. Developed using the Klarity™ computational biology platform, K757 is being evaluated for the treatment of obesity and type 2 diabetes, both as a monotherapy and in combination with K-833, a GPR119 agonist.
