KA-2887-LD38 is a novel bispecific antibody-drug conjugate (ADC) designed to target both Epidermal Growth Factor Receptor (EGFR) and Programmed Death-Ligand 1 (PD-L1). Developed by Escugen, the drug utilizes their proprietary LD-38 ADC platform, which features a topoisomerase II (TopII) inhibitor payload conjugated via a highly hydrophilic, stable, cleavable linker to a bispecific antibody (KA-2887). The antibody component was discovered using a common light chain platform and follows a conventional IgG1 structure. KA-2887-LD38 is engineered to treat solid tumors by exploiting the frequent co-expression of EGFR and PD-L1 in malignant tissues, which is rare in normal cells, thereby reducing 'on-target/off-tumor' toxicities. Preclinical studies have demonstrated potent tumor growth inhibition and strong bystander killing activity in models of non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma, and esophageal squamous cell carcinoma.
