KA-5484 is a humanized bispecific monoclonal antibody (BsAb) designed to simultaneously target OX40 ligand (OX40L) and tumor necrosis factor alpha (TNFα). Developed for the treatment of autoimmune and inflammatory disorders such as rheumatoid arthritis, it aims to provide synergistic immune modulation by concurrently blocking T cell co-stimulation and chronic inflammatory signaling. The molecule is engineered with a humanized anti-OX40L antibody (derived from the murine clone KD-0025) and an anti-TNFα single-chain variable fragment (scFv) based on the adalimumab sequence. It utilizes a human IgG4 backbone containing the S228P mutation to minimize Fcγ receptor binding and the YTE mutation to optimize pharmacokinetic properties. In preclinical studies using collagen antibody-induced arthritis (CAIA) mouse models, KA-5484 demonstrated potent dual-target engagement and superior efficacy in reducing joint inflammation and clinical arthritis scores compared to benchmark bispecific antibodies.
