KAL2DIOL is a pharmaceutical formulation of calcifediol (25-hydroxyvitamin D3) developed by Faes Farma for the treatment of vitamin D deficiency. Calcifediol is a pre-hormone and the direct metabolic precursor to calcitriol, the active form of vitamin D. By providing calcifediol directly, the drug bypasses the initial hepatic 25-hydroxylation step required for cholecalciferol (vitamin D3) activation. This mechanism allows for a more rapid and predictable increase in serum 25-hydroxyvitamin D levels, which is particularly advantageous in patients with hepatic impairment, malabsorption syndromes, or obesity. Formulated as soft gelatin capsules, KAL2DIOL serves as the substrate for renal 1-alpha-hydroxylase to produce calcitriol, which subsequently binds to the vitamin D receptor (VDR) to regulate calcium and phosphate homeostasis and support bone mineralization.
