Kallikreins are a subgroup of serine proteases that play a key role in the kallikrein-kinin system (KKS), which is involved in the regulation of blood pressure, inflammation, coagulation, and pain. Kallikreins act by cleaving kininogen substrates to release kinins such as bradykinin. There are two main types in humans: plasma kallikrein (KLKB1) and tissue kallikreins (KLKs 1–15). Plasma kallikrein circulates as an inactive precursor (prekallikrein) and is activated during inflammatory or coagulation processes. Tissue-type kallikreins have diverse physiological roles and are implicated in cardiovascular protection, wound healing, cancer progression (as biomarkers), and other pathologies[1][2][3][4][5][7][9]. Recombinant human tissue-type kallikrein has been used therapeutically for microcirculatory disorders such as acute ischemic stroke and diabetic complications[3].
