Overview

Kamuvudine-8 (K8) is a small molecule drug developed as a first-in-class dual inhibitor of inflammasomes and reverse transcriptase. It is administered via a sustained-release intravitreal implant specifically designed for retinal delivery. The primary mechanism of action involves blocking inflammasome activation triggered by multiple toxic pathways—including complement activation, amyloid beta accumulation, iron overload, retrotransposons—thereby inhibiting the downstream inflammatory processes that lead to cell death in degenerative diseases such as geographic atrophy (GA) secondary to age-related macular degeneration (AMD). Preclinical studies have also shown potential applications in neuroinflammatory diseases like Alzheimer’s disease and multiple sclerosis. The drug is being developed by Inflammasome Therapeutics and has reached Phase 2 clinical trials for GA/AMD and Phase 1 for diabetic macular edema[1][2][3][4][6][7].
