Drug intelligence / Profile preview
Kamuvudine K8 (also known as K8 or Kamzinoflast) is an investigational small-molecule dual inflammasome inhibitor developed by Inflammasome Therapeutics. Derived from the nucleoside reverse transcriptase inhibitor (NRTI) scaffold, Kamuvudine K8 is engineered to retain anti-inflammasome activity while reducing antiviral activity and mitochondrial toxicity. It targets the NLRP3 and NLRC4 inflammasome pathways, blocking activation triggered by multiple toxic drivers such as complement, retrotransposons, and oxidative stress. Delivered as a bioerodible, sustained-release intravitreal implant administered once every three months directly into the back of the eye, Kamuvudine K8 is primarily being developed for geographic atrophy (GA), an advanced form of dry age-related macular degeneration (AMD). In Phase 2 clinical trials, the 0.7 mg dose of K8 demonstrated a 54% reduction in GA lesion growth with a favorable safety profile, and a global Phase 3 pivotal program is planned.
