Kapiglucagon is a proprietary, water-soluble peptide prodrug of glucagon developed by ImmuPharma for the treatment of type 1 diabetes. It is specifically designed to overcome the stability and solubility limitations of native glucagon, which tends to aggregate in solution, making it historically difficult to use in continuous delivery systems. By temporarily masking key structural elements of the peptide through a proprietary prodrug approach, Kapiglucagon maintains high stability in aqueous formulations. Upon subcutaneous administration, the molecule regenerates native glucagon in vivo, which then acts as an agonist at the glucagon receptor to stimulate glucose release. The drug is primarily intended for use in next-generation dual-hormone artificial pancreas devices and is being advanced via the FDA's 505(b)(2) regulatory pathway.
