KAYO-1609 is a first-in-class, orally administered small molecule inhibitor of the retinoid pathway, developed by KayoThera. It is designed to inhibit retinoid synthesis in immune and tumor cells, thereby enhancing T-cell mediated killing of solid tumors. The drug targets genetically defined cancers that exploit the immunosuppressive effects of retinoids, including pancreatic, stomach, breast cancers, melanoma, certain sarcomas, and T-cell acute lymphoblastic leukemia (T-ALL). Unlike previous attempts at targeting the retinoid pathway—which failed due to toxicity—KAYO-1609 has demonstrated an excellent safety profile in preclinical studies across multiple species with no dose-limiting toxicities observed. Its mechanism involves blocking the formation of ligands for the retinoid nuclear receptor pathway. As of early 2024, KAYO-1609 has completed IND-enabling studies and is expected to enter clinical trials for late-stage and metastatic cancers[1][2][3][5][6][7][8].
