Overview

KAYO-1732 is an orally available small-molecule inhibitor of aldehyde dehydrogenase 1A3 (ALDH1A3), developed by Kayothera. It acts by inhibiting retinoid synthesis in the pancreas, adipose tissue, and vasculature, thereby targeting the retinoid nuclear receptor pathway. This pathway is implicated in acute metabolic dysregulation and cancer-promoting effects. By blocking retinoid synthesis, KAYO-1732 aims to restore glucose and lipid control, showing promise as a disease-modifying therapy for cardiometabolic syndrome, type 2 diabetes, and other metabolic disorders. The drug is being positioned as a first-in-class oral therapy with potential to prevent or reverse diabetes and multiorgan degeneration associated with cardiometabolic diseases[1][2][3][5].
