KAZ954 is a monoclonal antibody developed as an antineoplastic agent for the treatment of solid tumors. It specifically targets and binds to Ectonucleoside triphosphate diphosphohydrolase 2 (ENTPD2), an ectonucleotidase expressed on various tumor cells, inhibiting its hydrolase activity. This inhibition prevents the conversion of adenosine triphosphate (ATP) to adenosine diphosphate (ADP), resulting in increased intratumoral ATP:ADP ratios and promoting immune activation within the tumor microenvironment. Preclinical studies have shown that blocking ENTPD2 with KAZ954 delays tumor growth and enhances immune cell infiltration, effects that are further augmented when combined with immune checkpoint inhibitors or additional blockade of the adenosine pathway. The drug was being developed by Novartis for advanced solid tumors but clinical development has been discontinued[1][2][3][6][7].
