KB-7898 is an orally available small molecule inhibitor of the lysine acetyltransferase (KAT) domain of p300, developed by Kronos Bio for the treatment of autoimmune diseases, with a primary focus on Sjögren’s disease. The drug works by inhibiting p300, a key cofactor for interferon regulatory factor 4 (IRF4), which plays a central role in immune responses across multiple cell types including B cells and T cells. Preclinical studies have shown that KB-7898 reduces antibody production in B cells and cytokine production in T cells, thereby modulating pro-inflammatory signaling pathways implicated in chronic inflammatory diseases. In animal models, such as collagen-induced arthritis rats, KB-7898 demonstrated significant reductions in inflammation and disease markers. There are currently no approved therapies targeting the underlying cause of Sjögren’s disease; thus, KB-7898 represents a novel approach to address this unmet need.
