KB-9558 is a preclinical small molecule drug developed by Kronos Bio that targets the lysine acetyltransferase (KAT) domain of p300, a critical cofactor in the IRF4 transcription regulatory network. By inhibiting p300’s KAT activity, KB-9558 downregulates IRF4 expression—a key driver of multiple myeloma cell survival and proliferation. Preclinical studies have shown that treatment with KB-9558 leads to rapid and potent suppression of IRF4, resulting in anti-tumor effects in multiple myeloma models. The drug is also being explored for HPV-driven tumors due to its ability to disrupt viral oncogene expression via p300 inhibition[1][2][3][5][6][7].
