Overview

KB295 is a synthetic glycan developed by Kaleido Biosciences as a Microbiome Metabolic Therapy (MMT™) for the treatment of ulcerative colitis. It is designed to modulate the gut microbiome by acting as a specific growth substrate for beneficial bacteria while depleting pathobionts. In clinical and ex vivo studies, KB295 has been shown to increase the production of short-chain fatty acids (SCFAs), such as acetate, propionate, and butyrate, and reduce the relative abundance of the Enterobacteriaceae family. The drug was evaluated in an exploratory, open-label clinical study (NCT04508413) in patients with mild-to-moderate ulcerative colitis, where it demonstrated safety, tolerability, and a reduction in inflammatory biomarkers like fecal calprotectin and lactoferrin. Development was halted following Kaleido Biosciences' cessation of operations in 2022.
