Overview

KB5359 is an orally administered, gut-restricted small molecule agonist of the G protein-coupled receptor 119 (GPR119), currently under development by Kallyope for the treatment of metabolic disorders. By targeting GPR119 receptors located on enteroendocrine cells (L-cells) in the gastrointestinal tract, KB5359 stimulates the postprandial release of several key metabolic hormones, including glucagon-like peptide-1 (GLP-1), glucose-dependent insulinotropic polypeptide (GIP), and peptide YY (PYY). This coordinated hormonal release promotes glucose-dependent insulin secretion and enhances satiety, offering a potential therapeutic approach for type 2 diabetes and obesity. The drug's gut-restricted profile is a strategic design choice intended to maximize local activation within the gut-brain axis while minimizing systemic exposure, thereby potentially reducing the risk of off-target effects and improving the overall safety profile compared to systemic GPR119 agonists.
