KB707 is a genetically modified, replication-defective, non-integrating herpes simplex virus type 1 (HSV-1) vector engineered to deliver genes encoding human interleukin-12 (IL-12) and interleukin-2 (IL-2) directly to tumors. The dual cytokine expression is designed to stimulate a robust anti-tumor immune response by promoting cell-mediated immunity and systemic immune-mediated tumor clearance. Two formulations are in development: an intratumoral injection for accessible solid tumors and an inhaled formulation for lung delivery. KB707 is being developed primarily for the treatment of advanced or metastatic solid tumors, including non-small cell lung cancer (NSCLC) and melanoma, particularly in patients who have relapsed or are refractory to anti-PD-(L)1 therapies. The drug has received FDA Fast Track designation for both inhaled and intratumoral routes[2][5][6][7][9].
