Overview

KBA1413 is a fully human monoclonal antibody developed by Kling Biotherapeutics. It was identified from a cured acute myeloid leukemia (AML) patient who remains in long-term remission following allogeneic hematopoietic stem cell transplantation. Using Kling’s proprietary B cell screening platform, the antibody was found to recognize a unique, previously undescribed sialylated epitope on CD43 (also known as JL1 antigen), which is present on AML and myelodysplastic syndrome (MDS) cells as well as several solid tumor types including melanoma and breast cancer. KBA1413 mediates tumor cell killing via triggering NK-mediated antibody-dependent cellular cytotoxicity (ADCC) against AML cells both in vitro and in vivo, suggesting it may have contributed to the graft-versus-leukemia effect observed in the original donor patient. The molecule has also been engineered into a bispecific T-cell engaging antibody format to enhance its therapeutic potential, showing potent cytotoxicity against primary AML cells in preclinical models. Its main indications under development are AML, MDS, and certain solid tumors[1][2][6][7].
