Overview

KBA651 is an engineered SIRPα-based antibody-drug conjugate (ADC) developed by K2B Therapeutics for the treatment of solid tumors, including metastatic colorectal cancer (mCRC), ovarian cancer (OC), and small cell lung cancer (SCLC). The molecule consists of an engineered Signal Regulatory Protein Alpha (SIRPα) domain conjugated to the cytotoxic microtubule inhibitor monomethyl auristatin E (MMAE). KBA651 targets the CD47/SIRPα axis by binding to CD47, a "don't eat me" signal frequently overexpressed on tumor cells. This binding allows for the selective delivery of the MMAE payload into the tumor cells, leading to cell death. By targeting CD47, KBA651 may also block the inhibitory signal to macrophages, potentially enhancing the anti-tumor immune response. The program is currently in the preclinical stage of development.
