Overview

KBA751 (also known as eSIRPα-DXd) is a preclinical-stage antibody-drug conjugate (ADC) being developed by K2B Therapeutics for the treatment of various solid tumors. The molecule consists of an engineered Signal Regulatory Protein Alpha (SIRPα) component conjugated to a DXd payload, which is a potent DNA topoisomerase I inhibitor. KBA751 is designed to leverage the SIRPα/CD47 axis to deliver its cytotoxic payload directly to tumor cells. By utilizing an engineered SIRPα scaffold, the drug aims to achieve targeted delivery while potentially modulating the immune-checkpoint interaction between CD47 on tumor cells and SIRPα on myeloid cells. It is currently undergoing preclinical evaluation for multiple tumor types.
