KBMD-G is a nucleoside reverse transcriptase inhibitor (NRTI) analog being investigated for the treatment of bone metastatic prostate cancer (BMPC). Developed by researchers at the University of South Florida, it belongs to a series of analogs (including KBMD-E and KBMD-H) that target oncogenic signaling pathways critical for cancer cell survival and interaction with the bone microenvironment. KBMD-G has been shown to inhibit the Stat3, NFκB, and JNK/c-Jun signaling pathways, leading to the induction of apoptosis and pyroptosis in prostate cancer cell lines like DU-145 and PC-3. Additionally, it modulates cytokine production by upregulating interleukin-18 (IL-18) and downregulating IL-8 and IL-6, thereby disrupting metastatic stimulating signals in the bone microenvironment.
