KBMD-H is an experimental small molecule nucleoside reverse transcriptase inhibitor (NRTI) analog being investigated for the treatment of bone metastatic prostate cancer (BMPC). Developed by researchers at the University of South Florida, KBMD-H works by disrupting critical oncogenic signaling pathways, specifically inhibiting Stat3, NFκB, and JNK/c-Jun signaling. This inhibition leads to the induction of apoptosis and pyroptosis in prostate cancer cells, such as the DU-145 and PC-3 lines, while sparing healthy osteoblasts. Furthermore, KBMD-H has been shown to modulate the bone microenvironment by downregulating pro-metastatic cytokines like IL-6 and IL-8 and upregulating IL-18, thereby hindering the establishment and progression of prostate cancer metastases in bone tissue.
