Overview

**KCEM1** is a novel dual agonist peptide drug targeting both the glucagon-like peptide-1 receptor (GLP-1R) and melanocortin-4 receptor (MC4R) for the treatment of obesity and metabolic disorders. The compound is a chimeric molecule that resembles the GLP-1 receptor agonist exendin-4 at its N-terminal end and α-melanocyte stimulating hormone (α-MSH), an MC4R agonist, at its C-terminal end. This dual-receptor targeting approach is designed to achieve weight loss and improved glucose tolerance while potentially reducing gastrointestinal side effects compared to single-receptor targeting drugs like semaglutide and tirzepatide. In preclinical studies using diet-induced obese rats, KCEM1 demonstrated significant weight reduction (approximately 14.5% after 7 weeks) comparable to semaglutide and tirzepatide, but with superior glycemic control and significantly lower blood glucose levels during glucose tolerance testing. The compound was specifically formulated to treat hypothalamic obesity in children, a genetic condition rather than one related to calorie intake. KCEM1 is being developed by researchers at Syracuse University, the University of Pennsylvania Perelman School of Medicine, and Seattle Children's Research Institute, with lead investigators Robert Doyle, Matthew Hayes, and Christian Roth. The drug is currently in preclinical development with plans to move toward human trials.
