Drug intelligence / Profile preview
KCF18 is an *in silico*-designed, multi-target anti-inflammatory peptide decoy that simultaneously blocks the binding of three major pro-inflammatory cytokines—tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β), and interleukin-6 (IL-6)—to their respective cognate receptors (TNFR1, IL-1R, and IL-6R). Derived from the complex structures of cytokine-receptor binding domains, KCF18 possesses amphiphilic properties with positively charged and hydrophobic residues, allowing it to act as a decoy and prevent receptor-ligand interactions. It has been investigated in preclinical models for various inflammatory conditions, including endotoxemia, sepsis, vascular inflammation, liver injury, lung injury, and bladder outlet obstruction (BOO)-induced bladder dysfunction and fibrosis. When loaded into a thermosensitive hydrogel, KCF18 can be released gradually to provide sustained cytokine blockade.
