Overview

KD-182 is a second-generation chimeric antigen receptor T-cell (CAR-T) therapy developed by Nanjing KAEDI Biotech (KAEDI Medical). It specifically targets Claudin 18.2 (CLDN18.2), a tight junction protein isoform that is highly expressed in various gastrointestinal cancers but has limited expression in normal tissues. A key feature of KD-182 is the incorporation of a human PD-1 gene knockout modification, which is designed to enhance the persistence and anti-tumor activity of the CAR-T cells within the immunosuppressive tumor microenvironment. The therapy is currently in the preclinical stage for the treatment of solid tumors, particularly gastrointestinal malignancies such as gastric, pancreatic, and esophageal cancers.
