KD019 + trastuzumab + mFOLFOX-6 is a combination therapy regimen being investigated for the treatment of HER2-positive metastatic or unresectable adenocarcinoma of the esophagus, gastroesophageal junction (GEJ), and stomach. The regimen consists of KD019 (tesevatinib), an oral multi-targeted tyrosine kinase inhibitor that blocks EGFR, HER2, Src, and VEGFR2; trastuzumab, a monoclonal antibody targeting the HER2 receptor; and mFOLFOX-6, a standard chemotherapy backbone comprising oxaliplatin, leucovorin, and 5-fluorouracil. This combination aims to provide comprehensive blockade of the HER2 pathway and other signaling pathways involved in tumor growth, angiogenesis, and metastasis. It is currently being evaluated in a Phase Ib dose-escalation study sponsored by NYU Langone Health.
