Overview

KD026 (also known as SLX 4090) is an investigational small molecule drug that acts as a selective inhibitor of microsomal triglyceride transfer protein (MTTP). It is designed to target intestinal enterocytes with minimal systemic distribution (<1%), aiming to avoid the hepatotoxicity associated with earlier MTP inhibitors. The primary mechanism of action is inhibition of MTTP, which plays a key role in the assembly and secretion of plasma lipoproteins containing apolipoprotein B. By inhibiting MTTP in the gut, KD026 reduces triglyceride absorption and improves glycemic control. The drug has been developed for metabolic diseases such as type 2 diabetes mellitus and hypertriglyceridemia; development for dyslipidemias and obesity has been discontinued[1][3][5][6]. Kadmon Holdings originated its development after initial work by Surface Logix.
