KD033 is a clinical-stage immunocytokine and bispecific fusion protein composed of a fully human, high-affinity anti-human Programmed Death Ligand 1 (PD-L1) IgG1 antibody fused to the human interleukin-15 (IL-15) and IL-15 receptor alpha sushi domain. This design enables targeted delivery of IL-15 activity to the tumor microenvironment, particularly in PD-L1-expressing tumors, aiming to stimulate both innate and adaptive immune responses against cancer cells. The anti-PD-L1 component blocks the PD-L1 immune checkpoint pathway, while the IL-15 moiety expands key tumor-fighting cell types such as natural killer (NK), natural killer T (NKT), and memory T cells without expanding immunosuppressive regulatory T cells. Preclinical studies have shown that KD033 can inhibit tumor growth in both PD-L1 positive and negative models, including those resistant to other checkpoint inhibitors. Developed initially by Kadmon and later by Sanofi following acquisition, KD033 has been evaluated in phase 1 clinical trials for advanced or metastatic solid tumors[1][3][4][5][6].
