Overview

KD035 is a fully human monoclonal antibody antagonist of vascular endothelial growth factor receptor 2 (VEGFR2) that was developed by Kadmon to inhibit VEGF-A- and VEGF-C-mediated activation of VEGFR2 signaling in order to block tumor angiogenesis in solid tumors. Structural and functional studies show that KD035 binds with high affinity to the extracellular domains 2 and 3 of VEGFR2, overlapping the ligand-binding interface and thereby preventing receptor activation by endogenous VEGF ligands and downstream pro‑angiogenic signaling pathways. Although it demonstrated potent VEGFR2 blockade in vitro, clinical development for oncology indications was subsequently discontinued.
