KD045 is an investigational, small‑molecule next‑generation inhibitor of Rho‑associated coiled‑coil kinases (ROCK), being developed primarily for the treatment of fibrotic diseases such as idiopathic pulmonary fibrosis and organ fibrosis in lung, kidney, and liver. As a pan‑ROCK inhibitor, KD045 is designed to block ROCK‑mediated signaling pathways that drive pro‑fibrotic gene transcription, collagen deposition, and fibroblast‑to‑myofibroblast differentiation, thereby attenuating fibrosis and stabilizing endothelial and epithelial barrier function in multiple in vivo models. Preclinical data presented by Kadmon have shown that KD045 reduces fibrosis and improves barrier integrity across renal, hepatic, and pulmonary fibrosis models, and the company has pursued IND‑enabling activities intending to move the compound into clinical development for fibrotic indications. Separately, an unrelated KD-045 program from Nanjing KAEDI Biotech has been described in external databases as a ROCK‑targeting construct within a CAR‑NK/gene‑therapy platform for cancer; however, that oncology program is reported as discontinued and has not advanced beyond preclinical development.
