KD061 is an orally available small molecule ferroptosis inducer that targets the Iron Sulfur Cluster Assembly 2 (ISCA2) protein. Developed by Kuda Therapeutics, KD061 inhibits ISCA2, a component of the late mitochondrial iron-sulfur cluster (L-ISC) assembly complex, which triggers an iron starvation response leading to iron overload and lipid peroxidation-mediated cell death. Furthermore, ISCA2 inhibition by KD061 blocks the iron-responsive element (IRE)-dependent translation of HIF-2α and HIF-1α. Preclinical studies have demonstrated significant anti-tumor activity in models of clear cell renal cell carcinoma (ccRCC), melanoma, and clear cell ovarian cancer, with a favorable safety profile.
