KD3-IFN is an oncolytic adenovirus vector that has been engineered to selectively target and kill cancer cells. It contains mutations in the E1A region that prevent binding to cellular proteins p300/CBP and pRb, making it cancer-specific. The vector overexpresses the Adenovirus Death Protein (ADP) and expresses human interferon-α (IFN-α) late in infection, enhancing its antitumor activity. IFN-α has antiproliferative, cytotoxic, antiangiogenic, and immunomodulating effects, contributing to its anticancer efficacy. The combination of KD3-IFN with tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) further enhances antitumor activity through a four-pronged approach: oncolysis, ADP overexpression, IFN-α-mediated immunotherapy, and TRAIL-induced apoptosis. This combination has shown superior antitumor activity in animal models.
