KDM1 siRNA is a small interfering RNA (siRNA) designed to silence the expression of lysine-specific demethylase 1 (KDM1, also known as LSD1). KDM1 is an epigenetic enzyme that plays a critical role in histone modification by removing methyl groups from lysine 4 and lysine 9 of histone H3 (H3K4 and H3K9), as well as non-histone proteins like p53. In preclinical research, particularly in gliomas, silencing KDM1 with siRNA has been shown to inhibit cell proliferation and colony formation. Mechanistically, the reduction of KDM1 levels promotes the acetylation of p53 and the subsequent upregulation of its pro-apoptotic target genes, such as p21 and PUMA. Furthermore, KDM1 silencing is associated with increased levels of H3K4-me2 and H3K9-Ac modifications, leading to the induction of apoptosis and the reduction of stemness markers like CD133 and nestin in glioblastoma cells.
