KDM6B shRNA is a research-grade genetic tool used to silence the expression of the KDM6B gene (also known as JMJD3), which encodes a histone H3K27 demethylase. By utilizing the RNA interference (RNAi) pathway, shRNA constructs targeting KDM6B lead to the degradation of KDM6B mRNA, thereby reducing the levels of the KDM6B protein. This reduction results in an accumulation of tri-methylated lysine 27 on histone H3 (H3K27me3), an epigenetic modification associated with transcriptional repression. In preclinical research, KDM6B shRNA is frequently employed to study the role of this demethylase in various pathologies, including pancreatic cancer, multiple myeloma, and inflammatory conditions. For example, studies have shown that KDM6B knockdown can promote pancreatic cancer progression by downregulating the tumor suppressor C/EBPα. These shRNA tools are typically delivered using viral vectors such as lentivirus or adeno-associated virus (AAV) and are primarily used for target validation and mechanistic exploration in laboratory settings.
