Overview

Vividion Therapeutics is developing an orally bioavailable small molecule KEAP1 inhibitor for the treatment of inflammatory bowel disease (IBD) and other autoimmune diseases. Discovered via a proprietary covalent chemoproteomics platform, the compound works by inhibiting Kelch-like ECH-associated protein 1 (KEAP1). This inhibition prevents the ubiquitination and degradation of Nuclear factor erythroid 2-related factor 2 (NRF2), allowing NRF2 to translocate to the nucleus and upregulate the expression of antioxidant and anti-inflammatory genes. Preclinical studies have demonstrated that this program can significantly inhibit inflammatory cytokines and chemokines while promoting epithelial healing, aiming to provide a disease-modifying effect in chronic inflammatory conditions.
