Overview

Keliximab (CE9.1) is a primatized monoclonal antibody directed against the CD4 antigen on T-lymphocytes. Developed by IDEC Pharmaceuticals (now Biogen), it was created using "primatization" technology, which combines macaque variable regions with human constant regions (specifically IgG1 heavy chains and kappa light chains) to reduce immunogenicity while maintaining high binding affinity. Keliximab acts as a CD4 antagonist, inhibiting T-cell activation and potentially inducing the depletion of CD4+ T-lymphocytes. It was primarily investigated for the treatment of chronic severe asthma and rheumatoid arthritis, though clinical development was eventually discontinued. In the context of biotechnology research, CE9.1 is frequently utilized as a structural isotype control for other primatized antibodies, such as lumiliximab (IDEC-152), due to its identical human constant region framework.
