Overview

Ketobemidone is a powerful synthetic opioid analgesic of the phenylpiperidine class, used primarily for the management of severe pain. Its analgesic potency is comparable to morphine. In addition to its action as an agonist at mu-, kappa-, and delta-opioid receptors, it also exhibits N-methyl-D-aspartate (NMDA) receptor antagonist properties, which may contribute to its effectiveness in certain types of pain that are resistant to other opioids. The drug undergoes hepatic metabolism mainly via glucuronidation and N-demethylation pathways, with possible involvement of CYP3A4 enzymes. It is marketed in several Scandinavian countries for severe pain but has been withdrawn from some markets such as Sweden[1][2][3][5].
