Overview

Ketodex is an investigational combination therapy consisting of intranasal dexmedetomidine and intranasal ketamine, currently being evaluated for procedural sedation and analgesia in pediatric patients. The combination leverages the synergistic effects of dexmedetomidine, a selective alpha-2 adrenergic agonist that provides sedation and analgesia without significant respiratory depression, and ketamine, a dissociative anesthetic that primarily acts as an NMDA receptor antagonist. Administered via a mucosal atomization device (MAD), Ketodex is designed to provide effective sedation for procedures such as the closed reduction of fractures and dislocations in children, thereby avoiding the pain and distress associated with intravenous catheter insertion. It is currently being studied in a multicenter, adaptive randomized controlled trial led by Dr. Naveen Poonai to determine optimal dosing and non-inferiority to standard intravenous ketamine.
