KEY-507-G is an investigational, lower-sodium oral solution formulation of oxybate (gamma-hydroxybutyrate) developed by Jazz Pharmaceuticals for the treatment of narcolepsy, specifically targeting cataplexy and excessive daytime sleepiness. As a specific formulation variant evaluated in early-stage development, KEY-507-G was compared against Xyrem (sodium oxybate) in a first-in-human Phase 1 pharmacokinetic and bioequivalence study. The drug acts as an agonist at GABA-B and gamma-hydroxybutyrate (GHB) receptors in the central nervous system. Although KEY-507-G demonstrated bioequivalence to Xyrem with a 50% reduction in sodium content, Jazz Pharmaceuticals ultimately prioritized the development of JZP-258 (Xywav), which offered a more substantial 92% reduction in sodium, leading to the termination of the KEY-507-G program.
