KeyGen-GPC3 is a therapeutic development program by Beijing KeyGen Biotech targeting Glypican-3 (GPC3), a cell-surface oncofetal antigen that is highly expressed in hepatocellular carcinoma (HCC) but absent in normal adult liver tissue. The program encompasses both a GPC3-targeting monoclonal antibody and an antibody-drug conjugate (ADC) derived from it. The monoclonal antibody is designed to bind specifically to GPC3-expressing tumor cells, potentially mediating anti-tumor effects through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC) or interference with GPC3-mediated signaling pathways. The ADC variant utilizes the antibody's high specificity to deliver a cytotoxic payload directly into the cancer cells upon internalization, thereby enhancing therapeutic efficacy while minimizing systemic toxicity. Currently in the preclinical stage, these assets are being developed as targeted therapies for the treatment of GPC3-positive hepatocellular carcinoma.
