Overview

KF19514 is a small molecule dual inhibitor of phosphodiesterase 4 (PDE4) and phosphodiesterase 1 (PDE1). It has demonstrated anti-inflammatory and bronchodilator properties in preclinical studies. Mechanistically, it inhibits PDE4 and PDE1 activity, leading to increased intracellular cAMP levels. This results in reduced inflammatory cell accumulation (notably eosinophils and neutrophils), inhibition of bronchoconstriction, suppression of airway hyperresponsiveness, and prevention of airway remodeling. KF19514 has shown efficacy in animal models for asthma by inhibiting both acute allergic responses and chronic airway inflammation/remodeling induced by antigen or platelet-activating factor exposure[1][2][3][5][6][8]. It also suppresses tumor necrosis factor-alpha production[3]. The drug has been studied via inhaled, oral, and intravenous routes.
