KFP-H019 is a small molecule inhibitor of Exportin-1 (XPO1), also known as Chromosome Region Maintenance 1 (CRM1). Developed by Nanjing Carephar Pharmaceutical (Jiangsu Carephar), it is being investigated for the treatment of advanced cancers. XPO1 is a key nuclear export protein responsible for transporting various cargo, including tumor suppressor proteins (e.g., p53, BRCA1) and growth regulators, from the nucleus to the cytoplasm. Overexpression of XPO1 in many cancers leads to the depletion of these suppressors from the nucleus, facilitating tumor growth and survival. KFP-H019 works by binding to XPO1 and blocking the export of these proteins, thereby restoring their nuclear function and inducing cell cycle arrest and apoptosis in malignant cells. The drug is currently in the preclinical or IND-enabling phase of development.
