Overview

KG2032 is a monoclonal antibody identified for its specific binding to acute myeloid leukemia (AML) cells in roughly half of patient samples, but does not bind to normal leukocytes other than B lymphocytes. Its epitope is a subset of HLA-DRB1 (major histocompatibility complex, class II, DR beta 1) molecules where the 86th amino acid is not aspartic acid. As a result, KG2032 recognizes AML cells from patients with this specific HLA-DRB1 variant, particularly relevant after allogeneic hematopoietic stem cell transplantation (allo-HCT) when donor and recipient HLA-DRB1 alleles differ. KG2032 has been used to engineer CAR T and CAR NK cells for targeted AML immunotherapy, showing strong anti-leukemic activity and minimal toxicity in mouse models. It is under preclinical development, with clinical trials planned for relapsed AML post-allo-HCT[1][2][3][4][5][6][7][9].
